  • AxoDen: An Algorithm for the Automated Quantification of Axonal Density in Defined Brain Regions | eNeuro
    The rodent brain contains 70,000,000+ neurons interconnected via complex axonal circuits with varying architectures. Neural pathologies are often associated with anatomical changes in these axonal projections and synaptic connections. Notably, axonal density variations of local and long-range projections increase or decrease as a function of the strengthening or weakening, respectively, of the information flow between brain regions. Traditionally, histological quantification of axonal inputs relied on assessing the fluorescence intensity in the brain region of interest. Despite yielding valuable insights, this conventional method is notably susceptible to background fluorescence, postacquisition adjustments, and inter-researcher variability. Additionally, it fails to account for nonuniform innervation across brain regions, thus overlooking critical data such as innervation percentages and axonal distribution patterns.   • Alpha-Synuclein Phosphomimetic Y39E and S129D Knock-In Mice Show Cytosolic Alpha-Synuclein Localization without Developing Neurodegeneration or Motor Deficits | eNeuro
    Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by motor and nonmotor symptoms. Its pathological hallmarks include the accumulation of misfolded alpha-synuclein (α-Syn) in Lewy bodies and Lewy neurites. Phosphorylation of α-Syn is a prominent feature of these inclusions, but its role in disease pathogenesis remains unclear. To identify the role of α-Syn phosphorylation in synucleinopathy, we generated two Snca knock-in (KI) mouse models carrying phosphomimetic mutations at SncaY39 or SncaS129 ( SncaY39E or SncaS129D ) which manipulated epitopes phosphorylated in the PD brain. Both SncaY39E and SncaS129D KI mice displayed increased α-Syn phosphorylation, enhanced oligomer formation, and a shift of α-Syn localization from membrane-bound to cytoplasm. However, neurodegeneration in the substantia nigra was not observed up to 24 months of age.   • Predictive Representations in Hippocampal and Prefrontal Hierarchies | Journal of Neuroscience
    As we navigate the world, we use learned representations of relational structures to explore and to reach goals. Studies of how relational knowledge enables inference and planning are typically conducted in controlled small-scale settings. It remains unclear, however, how people use stored knowledge in continuously unfolding navigation, e.g., walking long distances in a city. We hypothesized that multiscale predictive representations guide naturalistic navigation in humans, and these scales are organized along posterior-anterior prefrontal and hippocampal hierarchies. We conducted model-based representational similarity analyses of neuroimaging data collected while male and female participants navigated realistically long paths in virtual reality. We tested the pattern similarity of each point–along each path–to a weighted sum of its successor points within predictive horizons of different scales.
