    Research released today reveals new strategies for treating chronic pain, post-traumatic stress disorder, and alcohol abuse by targeting the brain’s cannabinoid system. Targeting cannabinoid receptors — which bind both natural neurotransmitters and the psychoactive ingredient in marijuana — may provide some of the therapeutic benefits of marijuana without the side effects. The findings were presented at Neuroscience 2016, the annual meeting of the Society for Neuroscience and the world’s largest source of emerging news about brain science and health.

    Alzheimer's disease (AD) has traditionally been associated with amyloid-β plaques, but growing evidence underscores the role of neuroinflammation in disease progression. The autoinflammatory hypothesis of AD suggests chronic immune dysfunction contributes to neuronal damage, making immune modulation a promising therapeutic strategy. Cannabidiol (CBD), a phytocannabinoid with anti-inflammatory properties, may offer therapeutic potential. This study investigates how CBD independently influences two key neuroinflammatory regulators in AD: the Indoleamine 2,3-dioxygenase (IDO) pathway and the cyclic GMP-AMP synthase (cGAS) pathway. Though mechanistically distinct, both shape CNS immune responses. Targeting these immune-metabolic axes provides a mechanistic alternative to amyloid- or tau-based approaches by addressing upstream drivers of neuroinflammation and immune dysregulation.   • Effects of Cannabidiol, Hypothermia, and Their Combination in Newborn Rats with Hypoxic-Ischemic Encephalopathy | eNeuro
    Therapeutic hypothermia is well established as a standard treatment for infants with hypoxic-ischemic (HI) encephalopathy but it is only partially effective. The potential for combination treatments to augment hypothermic neuroprotection has major relevance. Our aim was to assess the effects of treating newborn rats following HI injury with cannabidiol (CBD) at 0.1 or 1 mg/kg, i.p., in normothermic (37.5°C) and hypothermic (32.0°C) conditions, from 7 d of age (neonatal phase) to 37 d of age (juvenile phase). Placebo or CBD was administered at 0.5, 24, and 48 h after HI injury. Two sensorimotor (rotarod and cylinder rearing) and two cognitive (novel object recognition and T-maze) tests were conducted 30 d after HI. The extent of brain damage was determined by magnetic resonance imaging, histologic evaluation, magnetic resonance spectroscopy, amplitude-integrated electroencephalography, and Western blotting.   • High-THC Cannabis smoke impairs incidental memory capacity in spontaneous tests of novelty preference for objects and odors in male rats | eNeuro
    Working memory is an executive function that orchestrates the use of limited amounts of information, referred to as working memory capacity, in cognitive functions. Cannabis exposure impairs working memory in humans; however, it is unclear if Cannabis facilitates or impairs rodent working memory and working memory capacity. The conflicting literature in rodent models may be at least partly due to the use of drug exposure paradigms that do not closely mirror patterns of human Cannabis use. Here, we used an incidental memory capacity paradigm where a novelty preference is assessed after a short delay in spontaneous recognition-based tests. Either object or odor-based stimuli were used in test variations with sets of identical (IST) and different (DST) stimuli (3 or 6) for low- and high-memory loads, respectively.   • Abstract
    Effects of oral Δ-9-tetrahydrocannabinol (THC) on reflexive and voluntary saccades in humans.
    The physiological and anatomical underpinnings of the acute cognitive effects of cannbinoids in humans are poorly understood. Here, we investigated acute cerebral effects of THC in 12 healthy human volunteers. Subjects received 10 mg oral Δ-9-THC (Marinol) and were tested with saccadic paradigms during a period of peak subjective intoxication and peak plasma levels of THC two hours after intake. Compared to baseline testing, no change in accuracy of reflexive saccades was found. However, memory-guided saccades showed a significant increase in variable tageting errors (gain variability: 0.24 vs. 0.19, p = 0.02) and a significant increase in erroneous anticipatory saccades during the memory delay (12.0% vs. 4.3%, p = 0.01). Likewise, a significant increase in antisaccade errors was found (13.2% vs 5.8%, p = 0.008). For all types of saccades, changes in latencies were subtle.
